Influence of fibroblasts conditioned medium (FCM) in the Thyroid Hormone‐induced cardiomyocytic hypertrophy

Thyroid hormone (TH) is a potent factor able to induce cardiomyocytic hypertrophy in vitro. Little is know about the contribution of fibroblasts conditioned medium (FCM) in this process. We investigate the role of FCM to the development of cardiac hypertrophy induced by T3 in vitro. Methods Cells were obteined from neonatal ventricles of Wistar rats by colagenase/pancreatin method. Myocytes (Myo) and fibroblasts (Fib) were segregated using Percoll gradient. After Fib cultures had received serum‐free medium (SFM) for 24h, they were submitted to SFM, supplemented with T3 (FCMT3) or no (FCM) for 24h. After 24h the mediums were collected, supplemented with T3 (FCM+T3 or FCMT3+T3) or no (FCM or FCMT3) and used like FCM on Myo. ANF expression (hypertrophy marker) was analysed using RT‐PCR. Result T3 (249.4„b46.5) and FCM (169.8„b33.3) promoted Myo hypertrophy, expressed as % by increased ANF. Myo receiving FCM+T3 showed an increase on ANF (165.0„b16.1). Addition of T3 to FCM was not able to increase ANF. Myo treated with FCMT3 showed an increase on ANF, compared to control (162.5„b26.2). However, the addition of T3 on Fib was not able to accentuate the increase on ANF. Myo treated with FCMT3+T3 showed an increase on ANF, compared to control (170.1„b14.3). However, the Myo treated with FCMT3+T3 were not able to induce an increase on ANF. TH induces Myo hypertrophy independently of its action on Fib. Financial Support: FAPESP, CNPq
