Background It is unknow that molecular macheniam of heart failure, it is unclear intracellular calcium overloaded if it is main factor induceing heart failure. Objective The research were to examne intracellular calcium overloaded inducing heart failure by endothelin ‐1 (ET‐1). Method [Ca2+]i is measured by Fura‐2/AM, with 3000‐fluorescence spectrophotometer. ET‐1(37.5ug/kg)were inject into a vein. The myocardial contractility indexes were recorded by RM‐6000 eight‐channel polygraph along with the electrocardial signals. Which includes HR LVSP LVEDP ¡Àdt/dt max AP. Result When ET‐1 is in 20, 40, 60 nmol/L, the concentration of [Ca2+]i appeares a short and quick inceadse at first, then followed by a slow sustained one, and shows a dose dependence. Within 30 sec. [Ca2+]i from 170.2¡À5.5nmol/L to 525.3¡À16.3 nmol/L (p<0.01) then [Ca2+]i appeares a slow and sustained increase, for 5min., to 485.3¡À14.5nmol/L (p<0.01). With both verapamil 1¡Á10‐5 mol/L and ET‐1 60nmol/L, the concentration of [Ca2+]i, dose not increase. Without calcium in extracellular solution, the concentration of [Ca2+]i is still enhanced by ET‐1 60nmol/L, but it is not so obvious as containing calcium. Data were compared with control by AP from 11.99+/−2.5 to 0 (p<0.01), LVSP from 5.985+/−0.8 to 0.33 +/−0.5 (p<0.05), (LVEDP) from 0.1729+/−0.1 to 0.665+/−0.81(p<0.01) +dp/dt max from, 199.5+/−19.2 to 106+/−0.18 (p<0.01), ¨ Cdp/dt max from 146+/− 43.6 to 19.9+/−2.1(p<0.01) However HR increase from 365+/−50 to 550+/−68 (P<0.001) These result indicate high dose ET‐1(37.5ug/kg) induces heart of failure. Conclusion The results suggest that intracellular Calcium overloaded inducing heart failure.
