ACTIVITY CHANGES OF BETAADRENOCEPTORS AFTER ABBOPLEGISOL INDUCED TRACHEOPLEGIA IN RAT TRACHEAL GRAFTS

Functional activity of rat tracheal grafts was studied after tracheoplegia induced with Abboplegisol solution, a commercial product. The study used 24 Wistar rats weighing 175‐200 g. Contractile responses were measured from tracheal rings placed in an organ bath (20 ml). The composition of the Abboplegisol solution was as follows (g/L): calcium chloride dihydrate 17.6,magnesium chloride hexahydrate 325.29,potassium chloride 119.3,sodium chloride 643,the solution osmolarity was 304 mosm/L and the mean pH was 3.8 (3.5‐3.9). Two groups (n= 12 each)were designed for the experiments,in group I,normal tracheal function was evaluated after its excision,in group II tracheal function was studied after 3 hrs of Abboplegisol treatment. The histological study was performed by using a sweep‐transmission electronic microscope. The results are expresssed as means +/‐ SEM. Statistical significance between the sets of data was tested by two‐way analysis of variance. Significance between maximum responses and pD2 values (the negative logarithm of the concentration evoking 50 % of the maximum response) was calculated using the Student′s t test. Contractile function was maintained unchanged although the release of products derived from cycloxygenase,but not from nitric oxide,was altered. Relaxation mechanisms mediated by beta‐adrenoceptor is also significantly altered,perhaps at receptor level,since adenylate and guanylate systems are not affected.A possible postreceptorial change in the G‐protein system can not be discarded.
