Protective effects of vitamin C on adipocyte health and modulation of triacylglycerol accumulation

Obesity is characterized by the presence of excess body fat, and is associated with increased risk for a number of chronic and sometimes debilitating diseases. Despite a proliferation of “diets”, fad or otherwise, the incidence of obesity continues to rise in the US. There are many reasons for any diets to fail, including the inability to adhere permanently to behavioral/dietary changes. However, emerging evidence suggests that aberrations of adipocyte metabolic systems may play a role in the etiology and progression of obesity or in resistance to weight loss efforts. The present study investigated the effects of treatment with vitamin C on adipocyte viability and triacylglycerol accumulation in the 3T3‐L1 murine pre‐adipocytes cell line. 3T3‐L1 cells were grown in Dulbecco’s Modified Eagles Medium (DMEM). At near confluence, cells were differentiated into adipocytes in DMEM media with insulin (10 μg/ml), dexamethasone (0.5 μM) and 3‐isobutyl‐1‐methylxanthine (0.5 mM). Differentiation was confirmed by staining of lipid droplets with oil‐red‐o. Adipocytes were exposed to a strong oxidant challenge for one hour ( tert ‐butyl hydroperoxide, 0.50, 0.75, and 1mM) followed by vitamin C treatment (100 μM) or ethanol control. Lipid accumulation and cell viability were monitored for two weeks with continued vitamin C treatment and supplement of DMEM with oleic acid (400 μM). Results show that vitamin C treatment improved adipocyte viability and structural integrity following oxidant challenge. Also, vitamin C effectively attenuated triacylglycerol accumulation in 3T3‐L1 adipocytes, compared to controls.
