Impact of fructan consumption on intestinal ion and nutrient transport in Salmonella‐infected weanling dogs

Diarrhea‐associated morbidity and mortality following Salmonella typhimurium infection impacts both humans and animals. Fermentable fiber added to formula reduced symptoms of S. typhimurium infection in neonatal piglets. We hypothesized that fructan consumption enhances intestinal ion and nutrient transport following S. typhimurium infection in weanling dogs. Thirty 12‐week‐old hound‐cross dogs were randomized to consume one of the following diets: 1) no prebiotic (control); 2) 1% short‐chain fructooligosaccharides; or 3) 1% inulin. On d 14, dogs were further randomized to receive an oral gavage of S. typhimurium DT104 (5x10 8 CFU) or 0.9% saline (n = 5 per treatment). Ileum and colon were collected 48h post‐infection for assessment in modified Ussing chambers. Transmucosal resistance, a measure of passive ion transport, was not impacted by diet or infection. Transmucosal short circuit current, a measure of active ion transport, was significantly reduced (p=0.037) in infected animals consuming inulin. Among non‐infected groups, ileal Na + ‐dependent glucose transport was lower (p=0.038) in inulin versus control group. Colonic Na + ‐dependent glucose transport was higher (p=0.036) following infection. Neither serotonin‐ nor carbachol‐induced chloride secretion was impacted by diet or infection. In summary, inulin consumption reduced ileal glucose transport and colonic active ion transport.
