Background We had determinethat Ginseng panaxadiol saponin monomer RB1 possess the protective effectin myocardial ischaemia of raw. Objective The study is to examine if Rb1 block dysfunction of L‐type Ca2+ channel by endothelin‐1 from isolated myocardial cell. Metheod Ventricular cells are isolated by collagenase. A standard whole cell L‐type Ica current is recorded by patch clamp technique. hold ¨ C50mV and were show the relationship of current ¨ Cvoltage. Results The active voltage of recorded currentis at ¨ C30mV, Peak current isat ¨ C10mV,the reverse voltage is at ¨ C60mV. The peak current is ¨ C302.7+/−28.16pA as controal. L‐Ica is enhanced with ET‐1 50nmol/L and shows the dose dependence. The peak current is enhanced from −251¡À32.85 pA to −423.6¡À48.56 pA(P<0.01) . L‐type single Ca2+ channel current is recorded. With ET‐1 50nm/L, the open time is prolonged, from 0.718¡À0.001ms to 1.044¡À0.0042ms (p<0.001), the close time is shortend from 8.344¡À0.93ms to 5.247¡À0.9ms (p<0.01), and the openstate probabilities increase from 0.179¡À0.015 to 0.137¡À0.004(p<0.01). The antagonistic action of Rb1 were observed. The enhanced whole cell L‐type calcium current induced by ET‐1 decreases from−423.6¡À48.56 pA(P<0.01) to ¨ C222.3+/−23.25pA (p<0.01). Increases open time of single channel induced by ET‐1 is shortened from 1.044 +/− 0.0042ms to 0.403 +/−0.007ms (p<0.01) Conclusion RB1 possess effect of blocking dysfunction of Calcium channel by endothelin‐1.
