Our previous study has revealed that substance P (SP) involves in water balance‐related pathophysiological process through modulation of a water channel (aquaporin 3). To characterize the effect of SP on aquaporin 3, we are conducting in vitro experiment in respiratory epithelial cell line (A549). Our current data show that SP (10 −9 –10 −11 M) significantly down‐regulated aquaporin 3 mRNA expression in a concentration‐dependent manner during 4 h incubation. The significant change was detected as early as 30‐min after SP addition. Sar 9 Met(O 2 ) 11 ‐substance P (10 −9 –10 −11 M), an agonist of SP, further confirmed this in vitro efficacy, indicating that both temporal/concentrational patterns follow the same trends as those of SP. This finding suggests that SP may play a role in the regulation of water channels (Supported by ALA and U. S. Air Force).
