Isolation and mapping of a polymorphic DNA sequence (pMCT106) on chromosome 2 [D2S61]

SOURCE/DESCRIPTION; A 1.7 kb Mspl fragment from cosmid MCT106 isolated by an oligonucleotide (GTGTGTGTGTGTGTGTGTGT) (1) was subcloned into the AccI site of pUC18. POLYMORPHISM: PvuII identifies a three allele VNTR polymorphism with bands between 2.0 and 3.5 kb without a constant band. Mspl, TaqI, Rsal, PstI also identify the same polymorphism. FREQUENCY; 32 % hetarozygosity has been observed with PvuII in 114 unrelated Caucasians. NOT POLYMORPHIC FOR: none detected CHROMOSOMAL LOCALIZATION: pMCT106 has been assigned to chromosome 2 by multipoint linkage analysis(2) with loci (ACP, D2S1) known to span this region(3). MENDELIAN INHERITANCE: Co-dominant segregation of the PvuII RFLP was observed in 57 three generation families. PROBE AVAILABILITY: Freely available (contact Y.N.) or will be available from ATCC. OTHER COMMENTS: RFLPs were observed under normal hybridization and washing stringencies.
