In this paper, microorganism for high lipase productivity was screened before purification, immobilization for biodiesel synthesis. High lipase producing strains such as Candida, Bacillus, Aspergillus, Pseudomonas sp. were screened and selected from oil-polluted soils. Micro-organisms were cultured for about two days after the elution of 1-fold, among which the best showed higher hydrolysis ability on fatty acid glycerides. Lipase productivity was improved 10-fold by mutation methods of ultraviolet treatment, and the most suitable time for treatment was 30 s. Furthermore, NTG was utilized to strengthen this mutagenic effect before selecting four strains with higher lipase productivity. These micro-organisms were cultured at their best conditions, which resulted in high lipase production after one day fermentation in 50 mL container at 29 °C.
