Significance We report the discovery of functions of intrinsically disordered RNA-binding protein RBM14 in NHEJ. RBM14 and KU proteins are recruited to DSB sites and required for the generation of RNA:DNA hybrids in response to DSBs. Despite a considerable number of intrinsically disordered RNA-binding proteins being implicated in DNA damage response, little is known about their roles in DNA repair pathways. Our findings provide insight into understanding how DSBs are repaired at transcriptionally inactive genomic areas and how the RNA-binding protein RBM14 plays a role in the NHEJ process.
