Summary Sera from mice injected with pneumococcal Type III capsular poly‐saccharide were titrated for antibodies by passive haemagglutination and haemolysis. The pattern of serological reactions suggested the existence of two distinct antibodies of the same specificity, one active in haemagglutination and of relatively high affinity for antigen, the other active in haemolysis and of lower affinity. These antibodies were entirely of the macroglobulin type. Titres of haemagglutinating and haemolytic activity were followed through the responses to three doses of antigen; some conditions for establishing substantial secondary responses were related to the two antibodies formed, their pre‐injection levels and the amounts of antigen administered.
