Summary A pylorus and oesophageal ligated rat preparation has been used to test the acid stimulating properties of crude gastrin, pure commercial gastrin and the synthetic “gastrin‐like” pentapeptide ICI 50123 given as subcutaneous injections. Amounts of 5 μg. and 25 μg. of gastrin and pentapeptide respectively were detected above basal control levels and maximum secretion was achieved with doses of 20 and 100 μg. respectively, supramaximal does giving inhibition of acid secretion. These results indicate that the pylorus and oesophageal ligated rat may be a suitable preparation for the bioassay of gastrin and related peptides.
