Summary A method is described for the estimation of testosterone in urine. This is a double isotope dilution derivative procedure similar to the method previously described for the estimation of testosterone in peripheral plasma. Testosterone is found in three fractions from urine—in the free fraction extracted at neutral pH, in a pH 1 fraction in which testosterone is released after incubation with mineral acid at pH 1 at 40°, and following incubation with β ‐glucuronidase. Values from a series of normal males and females art; given as well as from a series of patients with a variety of endocrine disorders.
