and Ab40. The method comparison revealed that wild-type Ab42 concentrations from LC-MS/MS were greater than those reported by the Innogenetics ELISA (on average 2.6 fold greater). Conclusions: Using instrumentation common to hospital labs, we developed an automated method for quantitation of Ab peptides in CSF, inclusive of familial Ab variants. The method was validated following established clinical guidelines and a data analysis procedure was established to identify the presence of variant Ab peptide sequences without a priori knowledge of the isoform(s) present in specimens submitted for analysis.
