dose, and an extracellular proton buffer, guanidinoethyl sulphonate (GES) (NeuroReport 1993;4:1035) given orally, daily for five days, unequivocally exhibited significant facilitation of GF. Neither agent had any adverse effects on animals. Conclusions:A direct AQP-4 facilitator and extracellular proton buffer appear to be effective agents for GF facilitation. Since neither showed any adverse effects on animals, TGN-073 and GES represent strong candidates as preventive, and potentially curative, pharmacologic agents for AD. Clinical studies are now pending.
