these levels with commonly used AD biomarker, amyloid beta 1-42 (Ab1-42). Methods:The study included 20 patients with MCI and 20 elderly individuals without cognitive impairment. The CSF concentrations of MMP-9 and Ab1-42 were determined using ELISA method. We also assessed the diagnostic sensitivity and area under ROC curves (AUC) of MMP-9 and Ab1-42in differentiation between MCI patients and non-demented controls. Results:Concentrations of Ab1-42 in MCI group and normal controls were 642 pg/mL and 1090 pg/mL, respectively. The CSF levels of MMP-9 were significantly lower inMCI patients (Median 107 pg/mL, p<0.001) in comparison with elderly individuals without cognitive impairment (Median 447 pg/mL). Additionally, ratios of CSF levels of MMP-9 to Ab1-42 in MCI patients (0.15, p<0.001) were significantly lower than in control group (0.45). Moreover, the percentage of positive results for MMP-9 (95%) was higher than for Ab1-42 (60%), whereas AUCs were 0.985 for MMP-9 and 0.763 for Ab1-42. Conclusions:Our findings suggest that MMP-9 might play a role in the development of mild cognitive impairment and could be a good candidate for the diagnostic biomarker of the early stage of dementia, although this issue requires further elucidation.
