IC‐P‐049: Detailed structural analysis of the hypothalamus in behavioral variant frontotemporal dementia

and cognitive decline. Several recent studies have shown that Ab accumulation is associated with cerebral changes prior to the development of cognitive impairment. In this work, we report 2-year longitudinal changes in gray matter volume in cognitively healthy subjects with abnormal levels of CSF Ab. Methods:A total of 42 cognitively healthy subjects underwent a T1-weighted 3T MRI and lumbar puncture to obtain biomarkers levels in the CSF. Two years later, MRI was repeated on the same subjects. The sample was dichotomized into Ab+ (< 500 pg/mL; N1⁄411) and Absubjects ( 500 pg/mL; N1⁄431). Longitudinal volumetric changes in gray matter were compared between the two groups using the Voxel Based Morphometry implementation in SPM12. Statistical significance threshold was set to p<0.001 uncorrected for multiple comparisons and cluster size > 100 voxels. Results: Ab+ subjects displayed significant decrements in gray matter volume in the bilateral hippocampi, superior and middle temporal cortices, insulae, caudates, cingulate cortex, and precuneus as well as in the left pars opercularis.Conclusions:Our results support that Ab deposition is associated with cerebral structural changes prior to the development of cognitive impairment in AD.
