Background: Tau hyperphosphorylation and memory deficit are characteristic pathological changes of Alzheimer’s disease (AD), and protein phosphatases (PP) 2A plays a crucial role in these AD-like changes.We had previously reported that inhibition of PP2A by okadaic acid (OA) induces tau hyperphosphorylation and memory impairment of rats. However, how to rescue tau hyperphosphorylation is not available now. Methods: Morris water maze was used to test the memory retention of rats, sterological injection was used to deliver OA (0.2 mmol/L)to hippocampus, L-carnintine was administrated by drinking(50mg/d$rat, P.O.) . Western blot was used to detect the tau phosphorylation in hippocampal homogenate, immunohistochemistry to detect tau phosphorylation in hippocampal slice. Results: Pre-treatment of L-Carnitine (50mg/d$rat, P.O.) for 2 weeks efficiently improved the OA-induced spatial memory retention impairment of the rats; it also antagonized tau hyperphosphorylation at multiple AD sites, arrested the inhibition of PP2A and recover oxidative stress induced by OA. Conclusions: Our study gave the first in vivo evidence that L-Carnitine could protect the inhibition of PP2A and improve tau hyperphosphorylation and spatial memory deficit of the rats, suggesting that this chemical may be served as a candidate for arresting AD-like pathological and behavioral alterations.
