Analogs of APP 5-mer peptide were synthesized and the most active ones were selected. The present study is to investigate the neuroprotection effect of the most active analog 165 in vitro and vivo. Methods: The most active analog was selected in human neuroblastoma cells (SY5Y) in vivo. The cells were divided into 3 groups: normal control group, damaged group (A 23-35 10 mol/L), and analog 165-protected group(20 mol/L). The function of analogs 165 was evaluated by counting the cell number, measuring the MTT metabolism, LDH leakage. The expression of P-CREB and Bcl-2 were detected by Westernblotting. In addition, the effect of analog 165 is also observed in STZ-induced diabetic mice. Morris water maze test was used to assess the learning and memory ability. Some survival and death related protein in hippocampal neurons were observed by immunohistochemistry and Western-blotting. At last, we observed the ultrastructural change. Conclusions: The experiments both in vivo and in vitro suggest that Analog 165 is an active fragment and have neuroprotective effect.
