no drug, fluoxetine, or D-serine in combination with either normal vision or 1 week of monocular deprivation. We used Western blotting to quantify expression of the glutamate receptor anchoring protein PSD-95, and GABAA receptor anchoring protein Gephyrin. We determined the PSD-95 to Gephyrin balance because their interactions regulate the number of E–I synapses and physiological E–I balance. We found that monocular deprivation alone caused a large shift towards excitation. While neither drug altered the E–I balance both drugs affected the shift driven by monocular deprivation. Fluoxetine decreased the magnitude of E–I shift and D-serine reversed it completely in favor of inhibition. Together, the results show that critical period-like plasticity reinstated by fluoxetine or D-serine in adult visual cortex uses different synaptic mechanisms to alter the experience-dependent E–I balance.
