AP2 as linker for clathrin to the endocytic membrane. We find that knock-down of the subunit AP2m1 by an antisense-morpholino based approach leads to a down-regulation of otx2 expression as well as an up-regulation of wnt/ -catenin target gene expression such as lef1. These data suggest an increase in wnt/ -catenin signalling possibly by an expansion of the efficiency range. In vivo imaging of the endocytic process of wnt8-GFP at early gastrulation stage, however, does not reveal any differences of the Wnt8 uptake-rate in responding cells judged by quantification of confocal images. Therefore we are presently testing other possibilities of an AP2m1 mediated regulation of the wnt/ -catenin signaling pathway.
