The purpose of our work is to identify the origin and mechanism of generation of neuronal diversity in the ventral telencephalon with a particular focus on inhibitory interneurons. We use Cre-lox fate mapping in transgenic mice to label specific populations of neural precursor cells and all their differentiated progeny in the embryonic telencephalon. We have previously taken this approach to identify the origin and heterogeneity of glial cells during development (Kessaris et al., 2006). We demonstrated that the majority of neuroepithelial precursors in the telencephalon generate oligodendrocytes that are functionally redundant; when one population is eliminated the rest of the cells make up for the loss (Kessaris et al., 2006). We are now taking a similar approach to address the question of origin and heterogeneity of telencephalic interneurons.
