the present study, we identify a new DRE in the second intron of the dopamine D2 receptor gene that binds to Freud-1 in vitro. Using chromatin immunoprecipitation we demonstrate an in vivo interaction between endogenous Freud-1 and the DRE elements in both 5-HT1A and dopamine-D2 genes. Knockdown experiments using siRNA of Freud-1 demonstrate up-regulation of dopamine-D2 RNA, particularly in cells that express low levels of the receptor. We conclude that Freud-1 negatively regulates both 5-HT1A and dopamine-D2 receptor expression, and hence may influence the development and activity of serotonin and dopamine systems. Further studies will identify the importance of Freud-1 in developmental regulation of 5-HT1A and dopamine-D2 genes and how Freud-1 induced regulation leads to developmental disorders, such as mental retardation, autism, schizophrenia or depression.
