In the CNS of the mouse embryo. L)M-20 was flrsr dctccred at E9.S in the latcrobasal plate of the dicncephalon. At E12.5, this lmtal rerr~lory extended rostratly to the ventral hypothalamus. Caudally, DM20” cells appeared in the latero-ventral plare of the rhombenccphalon. At E14.S, the number of DM-20+ cells in the rhombcnccphalon had Increased and extended all along the spinal cord. From E9.5 ull El4.5. no DM-20 signal W:I’ detected !n the mcscncephalon. Bctwccn E11.5 to birth, thi’ number of i)M-20+ cells ~ncrcascd and progrcsa~vcly colom~d ths future major \chltc smatter tracts.ln the chlckcn embryo. the “vcrall paucrn 01 cxprccs~on olL)M-20 was similar to the mouse. l’hc first D.WZiI+ ccII\; wcrc’ \e’cn at E?in the bnso-lateral plate I” ttrc mc\octlcnccl)~~~lllc sulcus end the rhombcnccphalon, but no Dk20 cells wcrc seen I” the inrcscnccphalic curvature. At E3, the UW-20 uanrcrq~t was dctcc~cd m the ventral hypothalamus From ES. the DM-20+ cells wcrc seen m rhc ventral rn:mtle layer, then rhcrc number p~mgrcss~ely mcrcased and they locah~cd mosrly m the future whltc matter tract. Double labclmg experiments falled LO show any colocalisatlon of DM-20 with ncuronal markers. In addition, cultnrr and grafting experiments showed that DA{-20+ ncur”ep~:hclial tcrrltories gcncratcd oligodcndrocytcs.
