Monoclonal antibodies (Mab's) that react with neuronal cell surface antigens were produced by fus ing X63-Ag 8.653 m y e l o m a cel ls w i th spleen cel ls f rom a mouse i m m u n i z e d w i t h adu l t rat s y n a p t i c p l a s m a m e m b r a n e s (SPM). ]n i n d i r e c t i m m u n o f l u o r e s c e n c e expe r imen ts three of these an t i -SPM Mab's, 3C5.59, 3G5.34, and 3G6.41 intensely stained the processes but only slightly stained the cell bodies of l ive g ranu le neurons cu l tu red f rom postnata l day 2-6 rat cerebel ls . Indi rect J m m u n o f l u o r e s c e n c e on cryostat sect ions of pos tna ta l days 1-18 and adult rat cerebella with Mab 3G5.34 show strong staining of the molecular layer. 3G6.41 stained both tbe developing moleculer layer and non-granule ce]l axons in the white matter of postnata l day 7 cerebe l lum. The externa l g ranu lar layer has l i t t le or no immunofluorescence in cryostat sections with either 3G6.41 or 3G5.34. In the adu]t cerebellum 3G6.41 did not stain the bulk of the mclecular layer, but rather stained only scattered fibers. 3G6.41 also strongly stained fibers in the white matter and surrounding the cell bodies of Purkinje cells. These data suggest that the antigen recogn ized by Nab 3G6.41 is found on granule cell axons only dur ing the early postnata l period. In cont rast Mab 3G5.34 recogn izes an ant igen that appears to be found on the granule cell axons throughout postnatal life.
