Nanocontrollers for In Vitro Drug Release Based on Core‐Sheath Encapsulation of Theophylline into Hydroxypropyl Methylcellulose Acetate Succinate Nanofibers

Linear adjustable efficient nanocontrollers that could respond to pH signals to trigger selective drug release were designed and discussed. They were fabricated by encapsulating theophylline into hydroxypropyl methylcellulose acetate succinate nanofibers through coaxial electrospinning. Theophylline was fixed in the core layer in an amorphous state. in vitro drug release behaviors were examined in simulated body fluid and tested by ultraviolet‐visible spectra. Expectedly, the nanocontrollers caused the drug to be autonomously released in weak alkaline solution rather than acid solution, with a sustained release over 12 hours. Thus, prior degrading or digesting in stomach can be avoided, and targeted drug releasing in the intestine can be realized. Moreover, the release rate was varied with regard to fiber diameter and drug content, indicating their adjustable property. The released performances were attributed to the synergistic effects of the chosen polymer and the core‐sheath structural advantages, for example, enteric solubility, drug encapsulation and large surface area. These results suggested that the designed nanocontrollers can serve as ideal candidates for a sustained targeted drug delivery system.
