Amylase activity in corn was determined with the Phadebas Amylase Test. Substrate concentration was varied between 1 and 6 tablets in a reaction volume of 10 ml. Reaction rate varied with substrate concentration according to a sigmoidal curve. The apparent kinetic parameters V max and K m were established. In the reaction volume of 10 ml 2 tablets of the Phadebas Amylase Test yielded a linear relationship between reaction rate and concentration of the enzyme extract. The substrate was attacked by a β‐amylase enzyme preparation as well. Amylase activity was found to be highest at 50 °C.
