Isolation, characterization and estimation of benzoxazinoid glycoside from seeds of Blepharis persica (Burm.f) O. Kuntze

Seeds of Blepharis persica , the extracts of which have various pharmacological activities, contain benzoxazinoid compounds. The present studies were aimed to isolate benzoxazinoid class of compound using chromatography, identify the isolated compound using spectroscopic methods, and development of a high‐performance liquid chromatography‐based analytical method for estimation of the constituent. The compound was isolated using HP‐20 and silica‐based column from n‐butanol fraction of Blepharis persica seeds and identified as blepharin from interpretation of spectra. An analytical method was developed using high‐performance liquid chromatography containing a photodiode‐array detector and validated as per ICH Q2(R1) guideline for estimation of blepharin. Blepharin was resolved using a C 18 column, whereas the optimized mobile phase was water (pH 8): acetonitrile (90:10, %v/v). The linearity was established between 0.5 to 10.00 μg/mL. Limit of detection and limit of quantification were 0.015 and 0.046 μg/mL, respectively. The recovery was in the range of 100.9‐103.9%. The method was found to be selective, precise, and robust too. The content of blepharin was found (0.708 ± 0.02)% w/w of the dried weight basis of plant extract.
