Peptides and proteins with molecular weights of ca 20 000 have been mass measured with accuracies of ±0.5 Da using electrospray ionization interfaced to a double‐focusing magnetic sector mass spectrometer. Factors affecting mass measurement accuracy such as resolution, peak profiles, calibration, data processing, and mass calculations are discussed.
