The hypothesis that an electrostatic effect of the buried aspartate side chain may contribute considerably to the extreme catalytic power of serine proteinases is presented. This statement is based on an analysis of environmental effects, due to amino acid residues in the vicinity of the Ser‐221…His‐64 couple and to hydration in subtilisin BPN'.
