Knowledge of the free magnesium ion content of intact cells is of fundamental biochemical interest. In collaboration with others, the author recently introduced a noninvasive 31 P NMR method for measuring the concentration of free Mg 2+ in intact cells and tissues. Applications of this method to the human normal and sickle red blood cells, frog skeletal muscle, and a metabolically highly active tumor cell have previously been described. This paper examines several points concerning the validity, advantages, and limitations of the NMR approach.
