In this study, fluorescence‐triggered two‐dimensional chromatography based on the application of a bromocoumarin tag facilitates the isolation of small molecule‐conjugated peptides prior to mass spectrometry. Profiling of a cysteinome with the use of a bromocoumarin tag demonstrates that two‐dimensional fluorescence‐based chromatography separation enables deep interrogation of select portions of cellular proteome.
