Two protein extraction procedures were tested in order to remove interfering compounds prior to 2‐DE of potato tubers. These methods using SDS lysis buffer and phenol‐phase extraction were compared regarding the quality of the resulting 2‐D gel. While the resolution of SDS extracts on semipreparative gels seems better, both methods lead to similar extraction yields and total number of spots. The procedures are complementary regarding the M r range of preferentially extracted proteins.
