Micro‐determination of oxalate in crude extracts of plant tissues

A convenient method is described for the accurate determination of small amounts of oxalate (at least as low as 0.02 μmol g −1 dry weight) in a number of plant tissues. Crude, acidic extracts of plant tissues are analysed without purification. The carbon dioxide evolved from the quantitative decarboxylation of oxalate by the enzyme oxalate decarboxylase is measured by gas chromatography. The enzyme was prepared from Collybia velutipes .
