Physical maps of the streptococcal bacteriophage A25 and C1 genomes

A physical map of the group A streptococcal bacteriophage A25 genome was constructed using data obtained from restriction enzyme digestion analysis of purified DNA. Phage A25 was 34.6 kb in size and was digested with several enzymes which generated the following number of fragments: Ava I (3), Eco RI (2), Hind III (4), Hae II (10), Hind II (22), Hpa I (11), and Pvu II (8). Restriction enzymes Bam H1, Pst I, and Sal I did not cleave phage A25 DNA. A physical map of the group C streptococcal phage C1 genome was also constructed. Phage C1 was 16.9 kb in size and was cleaved by the following enzymes to give the indicated number of fragments; Eco RI (8), Hae II (4), Hae III (10), Hind III (5), Hpa I (8), Pvu II (3). and Sal I (2). Restriction enzymes Ava I, Ava II, Bam H1, Eco RV, Hpa II, and Pst I did not cleave phage C1 DNA.
