Tritium labelled amitriptyline ( 1 ) of high specific activity was prepared. The tritium was incorporated by catalysed isotope exchange into the precursor dibenzosuberone, which had been previously converted into a cyclic ketal, in order to protect the oxo‐group against reduction. It was demonstrated by degradation that isotope exchange occurred exclusively in specified position.
