rac‐S ‐(2‐Acetoxy‐3‐hexadecyloxypropyl) thiophosphocholine ( 5 ), the isosteric and isopolar thiophosphate analog of Platelet Activating Factor (PAF), was synthesized from bis(2‐acetoxy‐3‐hexadecyloxypropyl) disulfide ( 12 ) or O,O ‐diethyl S (2‐acetoxy‐3‐hexadecyloxypropyl) thiophosphate ( 16 ) as starting materials by two independent „one‐pot” procedures in 65% and 20% overall yield, respectively.
