An ammonia gas sensor was used to determine 5′‐nucleotidase in serum through the ammonia produced from the coupled reactions of 5′‐nucleotidase and adenosine deaminase. Nonspecific hydrolysis of the substrate 5′‐adenosine monophosphate by serum alkaline phosphatase was corrected for by using the inhibitor concanavalin A. The present method is fast and simple, and offers considerable improvements over existing methods. Results obtained using the present method compare well with the spectrophotometric method and are not affected by sample turbidity.
