Many antitumor alkylating agents exert their cytotoxicity by targeting the O 6 ‐position of guanine to cause DNA damage. However, the therapeutic effects of the drugs can be significantly attenuated by O 6 ‐methylguanine‐methyltransferase (MGMT), which repairs the O 6 ‐alkylguanine lesions. In this paper, we describe a MGMT‐activated fluorescence turn‐on probe for the rapid no‐wash imaging of MGMT in living cells. We believed that the probe can be used as a prognostic indicator for tumor resistance to antitumor alkylating agents to provide a guide for therapeutic decisions.
