Standard methods inadequately address the presence of methemoglobin and methemalbumin that are indicative of severe intravascular hemolysis with rapid hemoglobin release. Here we present a novel second derivative fitting approach evaluating UV/VIS spectra for simultaneous quantitative analysis of oxyhemoglobin, methemoglobin, methemalbumin, and bilirubin, providing added diagnostic value when compared to standard spectrophotometric methods that typically underestimate the total free heme pigment content.
