The transcription initiation sites of the deo operon of Escherichia coli K12 have been determined in vitro and in vivo . Initiation of transcription occurs at two, tandemly arranged, homologous regions which are located 600 base pairs apart from each other. Evidence for a novel regulation mechanism of deo gene expression involving the deo R repressor is presented.
