A gel‐filtration procedure has been used to examine the effects of pH, ionic strength, enzyme concentration, and concentration of inorganic orthophosphate (P i ) on the molecular interaction between alkaline phosphatase and P i . At pH values above the isoelectric point of the enzyme, repulsion was demonstrated; this was reduced under those conditions favouring a reduction in the effective negative charge on either or both enzyme and phosphate molecules. In addition to providing information on the interaction between P i and enzyme, the study demonstrates that the gel‐filtration procedure can be used to study repulsion between molecules. Earlier it was shown that it could be used to study attraction.
