Effects of dibutyryl cyclic adenosine 3′: 5′ monophosphate and theophylline on 2‐deoxy‐D‐glucose and 2‐aminoisobutyric acid uptake by hamster embryo cells

The uptake of 2‐deoxy‐D‐glucose by hamster embryo cells was stimulated by preincubation with 1 mM dibutyryl cyclic adenosine 3′:5′ monophosphate (dibutyryl cAMP) and theophylline for 24 h. The increase in uptake depended on the concentration of dibutyryl cAMP and was accelerated by a combination of dibutyryl cAMP and theophylline. The increased uptake of 2‐deoxy‐D‐glucose was reversed by removal of these chemicals. Lineweaver‐Burk plots of the uptake indicated that these chemicals caused an increase in the maximum velocity of 2‐deoxy‐D‐glucose uptake but no detectable change in the apparent Km. Hexokinase was not activated by preincubation of cells with 1 mM dibutyryl cAMP and theophylline. The uptake of glucose in nine other cell lines, including those from rat and mouse embryos, was not stimulated by these chemicals. In contrast to the uptake of 2‐deoxy‐D‐glucose, that of 2‐aminoisobutyric acid was inhibited by these compounds.
