Ultracentrifugal studies demonstrate that cytotoxic methylhydrazine derivatives such as 1‐methyl‐2‐benzyl‐hydrazine break down nucleic acids much more than added hydrogen peroxide in solution of low content of iron ions. It is further shown that the methylhydrazine derivatives mentioned increase the reactivity of hydrogen peroxide towards deoxyribose. The conclusion is that the cytotoxic methylhydrazine derivatives not only produce hydrogen peroxide but that they also promote reactions between hydrogen peroxide and organic compounds. The latter effect is ascribed to the initiation of F ENTON chain reactions by the methylhydrazine derivatives or by the free radicals formed in the course of their autoxidation. The relevance of these results to the biological activity of the methylhydrazine derivatives has not yet been established.
