Lignanglucoside aus Podophyllum peltatum L . 7. Mitteilung über mitosehemmende Naturstoffe

From the rhizomes of american Podophyllum peltatum L . it was possible to isolate and characterize four lignan glucosides with anti‐mitotic activity: podophyllotoxin‐β‐ D ‐glucoside (IX), β‐peltatin‐β‐ D ‐glucoside (XI), 4′‐demethylpodophyllotoxin‐β‐ D ‐glucoside (X) and α‐peltatin‐β‐u‐glucoside (XII). The constitution of β‐peltatin‐β‐ D ‐ glucoside was deduced from the fact that enzymatic cleavage with emulsin yielded D ‐glucose and the already known β‐peltatin. α‐Peltatin‐β‐ D ‐glucoside, the first native glucoside of podophyllum t o be isolated in crystalline form, breaks down to D ‐glucose and α‐peltatin on hydrolysis with emulsin. The position of the sugar component in 4′‐ demethylpodophyllotoxin‐β‐ D ‐glucoside and in α‐peltatin‐β‐ D ‐glucoside could be determined by methylation to podophyllotoxin‐β‐ D ‐ glucoside and β‐peltatin‐β‐ D ‐glucoside. Distribution chromatography of the lignan glucosides on columns of silicagel proved a reliable analytical procedure. It could be used both as a test of purity and for assaying enriched fractions and crude plant extracts.
