(1) Ribonucleic acid has been extracted from isolated rat liver mitochondria by different methods (extraction carried out in perchloric acid, 10% sodium chloride or concentrated urea solution). The ratio of the bases was determined by chromatographic analysis after acid or alkaline hydrolysis of the nucleic acids. It is shown that the composition of the samples varies with the mode of extraction. Perchloric acid extracts furnished samples similar in composition to those found by other authors using whole liver ribonucleic acids. The preparations obtained with sodium chloride or urea solution showed a greater content of adenine and uracil. The significance of these results is discussed.
