The compressed conformations and degree of coiling of 1,ω‐diammonio‐oligo(ethylene glycol) (OEG) guests have been determined within the internal space of crystalline pyrene box host capsules. It was observed that the longest OEG chains have the tendency to fold in on themselves inside this confined medium. The phenomenon was observed in solution by NMR spectroscopy, and structural proof has been provided by single‐crystal X‐ray diffraction. The overall supramolecular spatial arrangement of the “pyrene box” system incorporating OEGs remains relatively unchanged compared with previously reported cages encapsulating alkane chains. However, some structural and dimensional variations/adaptations to accommodate coiled OEG guests of different sizes and shapes can be observed.
