A simple solution‐phase protocol for the synthesis of ceramide analogues from easily accessible enantiopure scaffolds is disclosed. The method relies on the use of nucleophilic thiolates or phenoxides and appropriate supported reagents or scavengers to give the target compounds in good overall yields. The method is easily adaptable to combinatorial protocols and also amenable to automated processes for the generation of small‐to‐medium‐sized libraries for further screening.(© Wiley‐VCH Verlag GmbH & Co.
