The cover picture shows the design and structures of 4′‐ C ‐alkylated thymidines. These modified nucleosides were synthesized and subsequently incorporated into oligonucleotides to probe inherent properties of the DNA double helix (left‐hand) side) and DNA polymerases in their interplay with their DNA substrates (right‐hand side).
