Approaches to the synthesis of purine derivatives bearing carbon substituents in positions 2, 6, or 8, mediated and/or catalysed by organometallic compounds and metals, are reviewed. Both the generation and the reactions of purinylorganometallic compounds are covered, as well as cross‐coupling and cycloaddition reactions between purines and organometallic compounds.
